Abemaciclib is used with hormone therapy for HR-positive, HER2-negative early breast cancer when there is a high risk of the cancer coming back after surgery.

It is also used for HR-positive, HER2-negative breast cancer that is locally advanced or has spread, either as a first treatment with an aromatase inhibitor or after earlier hormone therapy with fulvestrant. In some adults whose advanced HR-positive, HER2-negative breast cancer has worsened after hormone therapy and chemotherapy, abemaciclib may be used on its own.

Usual adult dose range: 50mg to 200mg twice daily; many breast cancer regimens start at 150mg twice daily with hormone therapy, or 200mg twice daily when used alone, with lower doses used if side effects occur.
