Palbociclib is used with an aromatase inhibitor as a first hormone-based treatment for HR-positive, HER2-negative breast cancer that is advanced or has spread.

It is also used with fulvestrant when HR-positive, HER2-negative advanced or metastatic breast cancer has worsened after earlier hormone therapy.

People who have not gone through menopause usually receive ovarian-suppression treatment as well.

Usual adult dose range: 75mg to 125mg once daily on days 1 to 21 of each 28-day cycle; the usual starting dose is 125mg once daily.
