Ponatinib is used for chronic myeloid leukaemia or Philadelphia chromosome-positive acute lymphoblastic leukaemia when the disease has a T315I mutation or when other tyrosine kinase inhibitors are not suitable.

It is also used with chemotherapy for adults with newly diagnosed Philadelphia chromosome-positive acute lymphoblastic leukaemia in some countries.

Usual adult dose range: 10mg to 45mg once daily, with the starting dose and later reduction based on the type of leukaemia, response, and risk of blood-vessel side effects.
